Granulocyte MCSF Human Recombinant produced in E.coli is a single non-glycosylated polypeptide chain approximately 14.6 kDa, containing 128 amino acids. GMCSF was initially characterized as a growth factor that can support the in vitro colony formation of granulocyte-macrophage progenitors. It is produced by a number of different cell types (including activated T cells, B cells, macrophages, mast cells, endothelial cells and fibroblasts) in response to cytokine or immune and inflammatory stimuli. Besides granulocyte-macrophage progenitors, GMCSF is also a growth factor for erythroid, megakaryocyte and eosinophil progenitors. On mature hematopoietic cells, GMCSF is a survival factor for and activates the effector functions of granulocytes, monocytes/macrophages and eosinophils. GMCSF has also been reported to have a functional role on non-hematopoietic cells. It can induce human endothelial cells to migrate and proliferate. Additionally, GMCSF can also stimulate the proliferation of a number of tumor cell lines, including osteogenic sarcoma, carcinoma and adenocarcinoma cell lines. GMCSF is species specific and human GMCSF has no biological effects on mouse cells.

Application Notes	< strong>Experiment Notes: GM-CSF quantitation was carried out by two independent methods1. UV spectroscopy at 280 nm using the absorbency value of 0.963 as the extinction coefficient for 0.1% (1mg/ml) solution. This value is calculated by the PC GEN computer analysis program of protein sequences (IntelliGenetics). 2. Analysis by RP-HPLC, using standard solution of GM-CSF as Reference Standard.

Storage & Handling

Storage	Store at 4°C for up to two weeks. For long term storage, aliquot and store at -20°C, avoid freeze/thaw cycles.
Buffer/Preservatives	GM-CSF was lyophilized after extensive dialysis against 2mM sodium phosphate buffer pH= 7.4 (with an error margin of +/-0.1).
