Rabbit polyclonal to SHC1.SHC-transforming protein 1 (SHC1) is a signaling adapter that couples activated growth factorreceptors to signaling pathways, such as the signaling cascade initiated by activated KIT andKITLG/SCF. The SHC1 gene encodes three main isoforms that differ in activities and subcellularlocations, including p46Shc, p52Shc and p66Shc. Isoforms p46Shc and p52Shc, oncephosphorylated, couple activated receptor tyrosine kinases to Ras via the recruient of theGRB2/SOS complex and are implicated in the cytoplasmic propagation of mitogenic signals. Isoformp66Shc does not mediate Ras activation, but is involved in signal transduction pathways that regulatethe cellular response to oxidative stress and life span. Isoform p66Shc acts as a downstream target ofthe tumor suppressor p53 and is indispensable for the ability of stress-activated p53 to induceelevation of intracellular oxidants, cytochrome c release and apoptosis.
